Alprostadil Cream As Effective As Injections For Erectile Dysfunction

ROBBINSVILLE, NJ -- May 11, 1998 -- Argentinian clinical study results show that NexMed Inc.’s Alprox-TD(TM) topical cream to treat erectile dysfunction (ED) proved as effective in producing targeted results as compared to intracavernous injections.

"The aim of our study was to evaluate the penile hemodynamic effect of a new topical formulation of alprostadil delivered to the corpus spongiosum through the glans skin," said Edgardo Becher, M.D., principal clinical investigator, professor of urology at the University of Buenos Aires and president of the Latin American Society for Impotence Research. "From the results of our study, we conclude that 500 mg of a 0.4 percent alprostadil cream incorporating the NexACT(TM) enhancer applied on the glans, produced a similar hemodynamic effect as determined by duplex ultrasonography as that seen following intracavernosal injection of alprostadil and sexual stimulation."

Alprostadil USP (prostaglandin E1, or PGE1) is recognised for treating ED. The active drug is readily available for the development of NexMed's Alprox-TD, a topical formulation incorporating the NexACT (acute transdermal drug delivery technology: specially-designed ingredients that promote the rapid penetration of the active ingredient through the skin and to the site of action and is the key to a rapid onset of action desired for a topical impotence product. The NexACT topical excipients are esters of fatty alcohols and amino acids and chemically designed to mimic the building blocks of the skin and be non-toxic and readily metabolized.

Numerous in vitro and in vivo penetration studies have shown that the NexACT enhancers result in the rapid delivery of alprostadil through the outer barrier layer of the skin as well as the tissue below the skin. Comparisons of Alprox-TD with a formulation of alprostadil without enhancers indicated that the control group had no effective cumulative penetration, while Alprox-TD had a significantly high penetration profile.

Pending regulatory approval, NexMed intends to market Alprox-TD in a single-dose package. The product has a shelf life of two years when refrigerated or one month at room temperature.
