| | | ![]() DG DISPATCH - APA: Olanzapine Effective for Juvenile Bipolar Disorder By Lara Pullen Special to DG News CHICAGO, IL -- May 18, 2000 -- Olanzapine is an effective monotherapy for juvenile bipolar disorder and improves patients' level of mania, depression and psychosis. At the 2000 Annual Meeting of the American Psychiatric Association, in Chicago, IL, Dr. Jean A. Frazier of Harvard University, Cambridge, MA, presented the results of a study designed to assess the safety and efficacy of the novel antipsychotic, olanzapine, after up to eight weeks of open-label treatment. Twenty-three bipolar disorder patients, ages five to 14, received olanzapine at doses between 2.5 and 20 mg/day. Efficacy was assessed using the Young Mania Rating Scale (YMRS) as the primary measure. Response was defined as > 30 percent improvement from baseline to endpoint in YMRS total score and a Clinical Global Impression - Bipolar Severity (CGI-BP) mania score < 3 (mild) at endpoint. Significant improvement (as assessed by YMRS) was seen by the first post-baseline visit at the end of week one and was sustained throughout the duration of the study. Olanzapine-treated patients also showed significant improvement in the Children's Depression Rating Scale (CDRS) total score. Treatment was well tolerated, with only one patient discontinuing their olanzapine treatment due to an adverse event (depression). The most significant adverse event was weight gain, which increased significantly during the course of the study.
